Hankinnan lyhyt kuvaus | Minimum functional requirements LC o UHPLC with maximum system pressure of at least 600 bar o Optionally biocompatibility. Wetted parts made of other than stainless steel o Column oven with capacity of at least one analytical column (200 mm) and one precolumn (50 mm) o Autosampler, which can be cooled down to 7°C ± 2°C o Degasser o Flow rate can be set between 0-4 ml/min being typically between 100 – 800 µl/min. Flow rate accuracy ≤ 5% and reproducibility RSD ≤ 0.5% o Injection volume between 0,1 – 50 µl. Injection reproducibility RSD ≤ 1% (based on peak area) o DAD or equivalent detector o Software for controlling and troubleshooting LC instrument. Compatibility of LC controlling software with MS software so that setting up and running sequences can be done via single UI MS o Ionization: ESI in positive and negative mode o Mass range at least m/z 50 – 3000 o Resolution (FWHM) at least 30000@ m/z 200 o Mass accuracy <3ppm o Possibility to perform top-down analysis of proteins o Possibility to analyse and quantitate small molecules and peptides o Possibility to perform MS/MS analysis in a quadrupole based collision cell o Dynamic range > 1000 o Data analysis software (can be several) that enables: Spectral library and database searches for small molecules Quantitation of small molecules Protein identification based on peptide MS2 data against sequence database and optionally against spectral library (bottom-up) Protein identification based on deconvolution of intact protein data and MS2 data and search against sequence database including intact protein mass information (top-down) Ideally combining previous two within a single piece of software |
