Hankinnan lyhyt kuvausThe University of Turku and Åbo Akademi University proposes to buy together a ThermoFisher ZipChip separation capillary electrophoresis (CZE) separation unit together with a Field Asymmetric Ion Mobility Separation unit (FAIMS). These units will be mounted (separately) to an existing ThermoFisher Lumos Orbitrap mass spectrometer. The Lumos is constructed in such a manner as to allow the analysis of intact proteins and to sequence them. Proteins cannot be reproducibly separated using the standard procedure of reverse-phase HPLC and so we need a CZE based separation. This allows separation of proteins that are modified (such as glycosylation, phosphorylation, deamidation etc.) which is essential for quality control of therapeutic proteins. The Universities also provide this type of service externally and the feature is therefore essential. In addition, FAIMS will allow to gain an extra dimension in peptide separation and by measuring the ion mobility increase the confidence levels of identifications. In addition, FAIMS opens up a novel approach to determining which drugs bind which protein. The two add-on separation techniques are controlled by software unique to the ThermoFisher Lumos Orbitrap mass spectrometer and will greatly expand the Universities repertoire of analytical techniques. FAIMS separates peptides and proteins by their shape and not mass by introducing a gas into the low-pressure region in front of the mass spectrometer after the ionisation of the molecules leaving the nanospray nozzle in the electrospray region. This instrumentation is completely unique to ThermoFisher. Other mass spectrometry companies have devices that can separate molecules by their shape using other physical methods, but all form an integral part of the mass spectrometer and cannot be fitted and controlled by a ThermoFisher instrument. The FAIMS unit is controlled by unique software produced by ThermoFisher that co-ordinates its function with the operation of the mass spectrometer. FAIMS is unique to ThermoFisher and there is no alternative. The ZipChip unit is a miniaturised capillary zone electrophoresis separation device constructed as a microchip in a special holder that interfaces only to the ThermoFisher electrospray source. Commercial CZE instrumentation has been available since the 1990’s. Now many companies offer CZE also in a chip format (such as PerkinElmer, Sciex, Agilent, Capilix, Isogen etc.). However, none of these other instruments can be controlled by the ThermoFisher software. This is essential since the main use is the ability to define the speed of separation to allow “peak parking”. The peptides and proteins are separated by applying a voltage across the capillary and it is possible to slow down and almost completely stop the passage of analytes by lowering the voltage allowing the mass spectrometer almost indefinite time to carry out MS/MS and MSn, analyses so greatly increasing coverage and depth of analysis. This requires online control of the separation that allows “intelligent control” of the separation. The alternative of using offline CZE separation is not feasible due to the extremely low analyte amounts and the fact that the throughput would be slowed down by around 50-fold. The ThermoFisher ZipChip is the only instrument format that can allow direct “intelligent control” of analyte separation since it requires direct control by the Orbitrap Instrument control language software. The ThermoFisher FAIMS and the ThermoFisher ZipChip are the the only instruments on the market which meets all the above mentioned necessary technical requirements to fulfill the upcoming needs of the Universities. According to the Act on Public Procurement and Concession Contracts (1397/2016) section 40 when only a certain supplier can implement the procurement for a technical reason, provided also that there are no reasonable alternatives or substitute solutions, and that the absence of competition is not due to an artificial narrowing of the terms and conditions of the procurement, the contract may be awarded only to a particular supplier, Contracting authorities may award public contracts by a direct award. Prior to making decision the Contracting authority has inspected the relevant markets and available products and after careful consideration found its' conclusion grounded. Since the ThermoFisher FAIMS and the ThermoFisher ZipChip are also expanssions to the existing Lumos Orbitrap mass spectrometer and change of supplier would result in procuring goods with different technical characteristics, causing incompatibility and disproportionately great technical difficulties in operation and maintenance. According to the Act on Public Procurement and Concession Contracts (1397/2016) section 41 A contracting entity may make a direct procurement when a procurement of goods to be made from an original supplier is an additional order that has been placed in order to partially replace or expand a previous delivery or installation. It shall be a condition that a change of supplier would result in procuring goods with different technical characteristics, causing incompatibility or disproportionately great technical difficulties in operation and maintenance. The duration of such agreements and renewable agreements may only exceed three years in exceptional circumstances.
