| Hankinnan lyhyt kuvaus | The University of Turku intends to buy a Chromium Connect automated single cell sequencing library preparation system. The Chromium Connect system is an integrated solution for automation of all key steps of single-cell RNA-sequencing sample preparation workflow, including single cell partitioning, molecular barcoding, and sequencing library preparation. The system utilizes a microfluidic reagent delivery method that enables performing thousands of micro-reactions in parallel. This allows automated partitioning of tens of thousands of single cells per run at high recovery rate (in the order of 60%) and generating barcoded cDNA and sequencing libraries from the cellular transcripts. Importantly, corresponding manual methods involve hundreds of pipetting steps and require at least one full working day, whereas the Chromium Connect system decreases the hands-on time to less than one hour per sample set. At the same time, the system significantly improves sample-to-sample consistency and minimizes technical variability due to operator effects. These are critical requirements for high-throughput core facility service use, that are not offered by any other currently available instruments. The Chromium Connect system is compatible with solutions for whole transcriptome analysis with a sensitivity of in the order of thousands of unique transcripts per cell, and a solution for multiplexed analysis of whole transcriptome and paired immune receptor sequences from T and B lymphocytes. These solutions are provided and fully supported by the instrument manufacturer (10x Genomics), including a proprietary pipeline form sequencing data processing, demultiplexing, and quality control. At the moment, no other instrument on the market is capable of performing these protocols in combination with fully automated and efficient single cell capture. The Chromium Connect is the only instrument on the market which meets all the above mentioned necessary technical requirements to fulfill the upcoming needs of the University. |
